Project A03 (E) - Mechanical characterization of integrin-dependent leukocyte adhesion

This project ended 06/2015

We will mechanically characterize integrin-specific adhesion forces using AFM and evaluate the modulation of integrin binding affinity and avidity through specific intracellular integrin adaptor proteins. Furthermore, we will characterize the influence of chemokines on cell polarity and on integrin adhesion forces at a single cell level. Constructing in parallel an enhanced-throughput force spectrometer, we will screen for additional modulators of regulating integrin-binding forces in lymphocytes and myeloid cells.
