Project A09 (E) - Protein sorting at the trans-Golgi network (TGN) and its impact on immune cell migration

Leukocyte trafficking in development, immune surveillance and inflammation relies on a plethora of secreted proteins such as cytokines, chemokines and matrix degrading enzymes. However the mechanisms controlling the export of these molecules from the Golgi apparatus remain largely unknown. Thus the goal of our project is to establish a link between protein sorting at the TGN and its relevance for leukocyte extravasation. To this end we will dissect a recently identified SPCA1/Cab45 dependent sorting process. Furthermore we will analyze the so far unknown pathway of MMP sorting in vitro and in vivo.
